Cathepsins, the lysosomal proteinases, are distributed in normal tissues and expressed in a variety of physiological and pathophysiological processes such as bone resorption, muscular dystrophy, arthritis and tumor invasion and metastasis.
They are synthesized as proenzymes which are processed to the active form by removal of proregion.
Mature cathepsins are localized in the lysosomes where they function in protein turnover but have also been shown to be capable of degrading ECM proteins. Higher levels of several cathepsins have been associated with thepathological destruction of the ECM leading to tumor metastasis, osteoporosis and rheumatoid arthritis.
